Binding characteristics of lectins involved in the trapping of nematodes by fungi.

چکیده
Seventeen saccharides were tested for their ability to bind to the trap lectins of three species of nematode-trapping fungi and prevent nematode capture. The lectin of Arthrobotrys conoides was found to be inhibited by alpha-d-glucose/d-mannose and similar saccharides. The lectins of Monacrosporium eudermatum and Monacrosporium rutgeriensis were inhibited by alpha-l-fucose and 2-deoxy-d-glucose, respectively. Human group O(H) erythrocytes agglutinated to traps of M. eudermatum but not A. conoides or M. rutgeriensis. There was no agglutination of group A or B to traps formed by all three fungi. Exposure of the traps to trypsin eliminated the ability to capture nematodes. The presence of d-glucose/d-mannose and l-fucose residues on the nematode cuticle was suggested through the use of commercially prepared lectin-peroxidase conjugates.

Screening of Soil and Sheep Faecal Samples for Predacious Fungi: Isolation and Characterization of the Nematode-Trapping Fungus Arthrobotrys oligospora
Over one-year period, 150 pasture soil samples and 138 sheep faecal samples, collected from different parts of Iran were screened for the presence of nematophagous fungi. The samples were cultured at 25ºC on chloramphenicol-2% water agar (CHF-WA) plates in the presence of Haemonchus contortus third stage larvae (L3) and checked over a two-month period for characteristic conidia, conidiophores a...
متن کاملPredatory behaviour of trapping fungi against srf mutants of Caenorhabditis elegans and different plant and animal parasitic nematodes.
The initial infection process of nematode-trapping fungi is based on an interaction between the trapping structure of the fungus and the surface of the nematode cuticle. A bioassay was designed to investigate the predatory response of several isolates of nematode-trapping fungi against 3 mutants of Caenorhabditis elegans (AT6, AT10 and CL261), which have been reported to differ in the reaction ...
متن کاملComparative Genomics and Transcriptomics Analyses Reveal Divergent Lifestyle Features of Nematode Endoparasitic Fungus Hirsutella minnesotensis
Hirsutella minnesotensis [Ophiocordycipitaceae (Hypocreales, Ascomycota)] is a dominant endoparasitic fungus by using conidia that adhere to and penetrate the secondary stage juveniles of soybean cyst nematode. Its genome was de novo sequenced and compared with five entomopathogenic fungi in the Hypocreales and three nematode-trapping fungi in the Orbiliales (Ascomycota). The genome of H. minne...
متن کاملTwo genes encoding fruit body lectins of Pleurotus cornucopiae: sequence similarity with the lectin of a nematode-trapping fungus.
Our previous studies on the fruit body lectin of Pleurotus cornucopiae revealed the existence of three isolectins, composed of two homodimers and one heterodimer of 16- and 15-kDa subunits. In this study, two genes encoding the lectins were cloned and characterized. Both genes encoded 144 amino acids and only 5 amino acids were different within the coding region, but the nucleotide sequences of...
متن کاملIsolation and Molecular Characterization of Indian Isolates of Nematode Trapping Fungi
Two nematode trapping fungi were isolated from aerated soil samples collected fron Anand district( Gujarat) India and identified on the basis of their culture –morphological characteristics and 18S rRNA gene sequencing as Arthrobotrys oligospora and Duddingtonia flagrans . These fungal isolates were assessed for their trapping efficiency further. Trapping structures such as ring like structures...
